NeOnc Technologies Holdings, Inc. (NASDAQ: NTHI) has announced positive results from its Phase 2a clinical trial of NEO100, a drug candidate for recurrent brain cancer, according to an update from Stonegate Capital Partners. The trial met its primary endpoint, with a six-month progression-free survival (PFS) rate of 48.9% as measured by RANO 2.0 using Kaplan-Meier estimation, compared to a pre-specified benchmark of 20% (p=0.0047). The median overall survival (OS) reached 26.09 months, and no major toxicities were reported.

While the PFS endpoint was a key measure, the survival signal is considered more important, as current salvage therapy for recurrent brain cancer typically yields only 6–9 months of survival, according to management. The positive readout moves NEO100 into a potential registrational program, with the company intending to request a Type B meeting with the FDA to discuss trial design and approval pathways. This regulatory clarity is seen as the next near-term catalyst for the company.

In addition to NEO100, NeOnc's second clinical program, NEO212, has gained regulatory momentum through Phase 2 CMC clearance and FDA feedback indicating a potential accelerated approval pathway. This broadens the investment case beyond a single asset, with NEO100 also being explored in meningioma and pediatric brain tumors, while NEO212 offers a differentiated approach.

The financial results for the second quarter of fiscal year 2026 were secondary to the clinical progress, with R&D expenses increasing to $2.6 million from $0.7 million year-over-year. This increase reflects the expanded development activities across the pipeline.

The implications of these results are significant for patients with recurrent brain cancer, who currently have limited treatment options. If NEO100 continues to show promise in a randomized study, it could offer a new, well-tolerated therapy that extends survival. For the industry, this highlights the potential of targeted therapies in challenging oncology indications.
